Histochemical Study of Pregnant Uterine Endometrium and Placental Villous.


Abstract
There has been an increasing interesting and extensive studies on the function of villi of placenta and pregnant uterus which are boundary tissue maintaining relation betwee mothers body and fetus. Recently, Wielenga, Deane, Label and Romney made various researches in the villi with the development of histochemistry. Author studied on the P.A.S. positive substance, nuclear acid, lipoid substance and phosphatase which were detected fron the villi and endothelium of 11 cases of 5-16 weeks¢¥ gestation, 14 cases of 17-36 weeks¢¥ gestation and 27 cases of full term gestation. The results are as follows :
1. The form of changes in the endothelium had been almost invariable and constant throughout the period of pregnancy. The structure of the villi of placenta exhibited marked changes according to the period of pregnancy. The trophoblast is consisted of two clearly defined cytotrophoblast at the early stage of pregnancy and its internal layer becomes discontinuous at the middle stage and internal and external layers are fused into one at the end stage. The supply of vessels of intervillous spaces are more increased and fibrisation becomes more marked in proportion as pregnancy proceeds.
2. The P.A.S. positive substance which are found in the cytoplasm of decidual cell of endothelium and uterine glandular epithelial cell are gycogen. It appears in the decidual celll mostly only at the end stage of pregnancy and is found generally throughout in the uterine? glandular epithelial cell.
The P.A.S. positive substance of Langhans cell of villi and interstitial cells are glucoprotein or mucoprotein and exist localizing only in those cytoplasm. But, judging from the degree of stain reaction, it can be said by no means that it is large in quantity. Meanwhile, the attitude of substance which exist in the Langhans cell is particularly worthy of note.
3. It appears that R.N.A. can be seen generally only in nucleolus and cytoplasm of pregnant ut-endometrium and villi of placenta, abundantly at the early and middle stage. The stain reaction of this substance appears strongly in the cytoplasm of ut-glandular epithelium and nucleolus of syncytial cell and weakly in other cells and can not be seen in the cytoplasm of interstitial cells throughout the pregnancy.
D.N.A. exist only in the nucleus of observed cell. This substance is characterized by the tendency of increasing in proportion as pregnancy proceed.
4. Cholesterol appears only in the cytoplasm of syncytial cell and most markedly at the middle stage of pregnancy.
Neutral fat, cholesterol, lipid were demonstrable only in the cytoplasm of other cells except for syncytial cell, such as decidual cell,ut-glandular cell, Langhans cell and interstitial cell. There was no noteworthy change in these substances throughout the pregnancy.
5. Alkaline phosphatase was demonstrable in the decidual cell ut-glandular cell syncytial cell and interstitial cell.
The activity of those enzyme, generally speaking, seems to have the tendency to be weakened, approaching to the end stage of pregnancy. The most noteworthy fact about this enzyme is that Langhans cell has. almost none of this substance.
Acid phosphatase was demonstrable only in the cyncytial cell and Langhans cell but very faintly. It is interesting to notice that this enzyme is definitely demonstrable only at the early and middle stage of pregnancy as it was also true in case of alkaline phosphatase.
